Remove barcodes from 3' end only

My barcode file is as follows:

NN,ATCGT:01_Fast_WT_1
NN,AGCTA:02_Fast_WT_2
NN,CGTAA:03_Fast_Mut_1
NN,CTAGA:04_Fast_Mut_2

However, I am encountering an error relating to the lack of non N characters in my 5' barcodes. My reads do not have barcodes on the 5' end, rather an NN UMI. What parameters could I use to demultiplex the above?

I'm afraid there will need to be at least 1 barcode at the 5' end. You could spin it around (pretend the forward read is reverse) and then use umitools or similar to extract the umi. But it's not ideal, sorry! Get Outlook for Android<https://aka.ms/AAb9ysg>
